APPLICATION OF POLIMERASE CHAIN REACTION FOR DETECTION OF Mycoplasma spp IN THE ROUTINE OF CELL CULTURES

The occurrence of Mycoplasma spp as contaminant of cell cultures in cell culture laboratories of the Escola de Veterinária da UFMG and of the Laboratório Nacional Agropecuário de Minas Gerais was obtained by the use of a fast DNA extraction protocol e by the use of PCR. The PCR sensitivity and specificity were evaluated. The search for Mycoplasmas DNA was done in 83 cell lines samples. We found infected cell lines in both laboratories. The quality of extracted DNA was verified using a beta-actin gene PCR. The Mycoplasmas’s PCR showed to be fast, sensitive and specific.
